Comparative Study of Plasminogen Activators in Cancers and Normal Mucosae of Human Urinary Bladder1

چکیده
We have developed a highly sensitive sandwich enzyme immunoassay for determination of urokinase-type plasminogen activator (u-PA) and tissue-type plasminogen activator (t-PA) antigen levels in extracts of human tissues. We determined antigen levels of PAs in extracts of 31 primary cancers and 15 normal mucosa! tissues of the urinary bladder using this method. U-PA antigen levels in extracts of bladder cancers were significantly higher than those in normal tissues (p < 0.005). U-PA antigen levels significantly increased as histolÃ3gica!grading of malig nancy advanced. There was no correlation between t-PA antigen level and malignancy. These results indicate that an increase of u-PA antigen level may be a parameter of malignant transformation and may play an important role in invasiveness of cancer.

متن کاملAntigenic Profile of Muscularis Mucosae and Muscularis Propria of the Urinary Bladder
Background: Differentiation between the muscularis mucosae (MM) and muscularispropria (MP) of the bladder remains challenging. Objective: To identify MM- and MP-specific antigens that could be of potential value for staging of urothelialcarcinomain a pilot study. Method: The expression of 12 protein antigens in 11 human bladder specimens was examined. There were 5 post radical cystectomy specim...

Background: Human plasminogen is a plasma glycoprotein synthesized mainly in the liver. Conversion of plasminogen to plasmin by plasminogen activators is a key event in the fibrinolytic system. In this study, we investigated the effects of two anti-human plasminogen monoclonal antibodies, A1D12 and MC2B8 on Glu-plasminogen activation in presence of u-PA, t-PA and streptokinase. Methods: Produci...
متن کاملPreparation of Plasminogen by Affinity Chromatography
Background: Plasminogen is one of the compounds derived from human plasma. Activation of plasminogen produces plasmin. Plasmin is able to lyse fibrinogen, fibrin, and some other human plasma proteins. The aim of the present work was to study the separation of human plasminogen by affinity chromatography using gel lysine Sepharose. Materials and Methods: Normal human plasma was used as the...
